Drug Class Proton pump inhibitor
Mechanism Inhibits gastric acid secretion by dose-dependent inhibition of proton pump of gastric parietal cells (blocks H+/K+ pump ATPase enzyme in parietal cells)
Typical Dose 20-40mg OD
Indications
  • GORD
  • dyspepsia
  • Oesophagitis
  • GI ulcers
  • H. pylori eradication
  • Zollinger-Ellison syndrome (gastrin-secreting tumour or hyperplasia of the islet cells in the pancreas)
Contra-indications

Caution in: 

  • Liver disease
  • Breast feeding/pregnancy
Side-effects
  • GI disturbances
  • Headache
  • Long term use associated with hypomagnesemia
Interactions Inhibits cytochrome P450 ↑ plasma concentration of phenytoin, warfarin
Elimination 80% renal, 20% faecal
Additional Notes May mask symptoms of gastric cancer
